Mycobacterium tuberculosis (MTB) is the most important and common pathogen causing tuberculosis (TB) in humans. It can invade various organ systems throughout the body, with lung infection being the most common infection. MTB is mainly spread through air, dust, and aerosol. A patient with a severe infection may suffer from hemoptysis as well as coughing and low-grade fever. Therefore, early detection and treatment of active TB is crucial. Bacillus Calmette-Guerin (BCG) is currently the only TB preventive vaccine approved for clinical use.

Main Steps of IVD for Mycobacterium Tuberculosis
Tuberculin test (PPD test). Based on the principle of delayed skin hypersensitivity, tuberculin is injected into the skin. Those with redness, swelling, and induration exceeding 5 mm after two or three days are considered positive.
IFN-γ release assay. After injection of tuberculosis-related antigen, the interferon content in the serum was quantitatively detected. Increased interferon indicates possible Mycobacterium tuberculosis infection.
DNA Testing. It mainly includes PCR, Xpert MTB/RIF test, 16S rRNA highly variable region sequence determination, and restriction length polymorphism analysis.
Direct smear staining examination. Smear directly from expectoration or smear after collecting bacteria, and stain with acid-fast stain.
Isolation culture. The sample is inoculated into a solid medium and cultured under suitable conditions for some time. If cauliflower-like colonies appear, the infection is determined.
